چکیده

Using mRNA differential display, we found that the gene for NAD(+)-dependent glycerol phosphate dehydrogenase (GPDH; EC 1.1.1.8) is induced in rat brain following seizure activity. Northern blot and in situ hybridization analysis confirmed the differential display results; they also showed, in a separate model of neuronal activation, that after thermal noxious stimulation of the hind-paws, a similar increase in GPDH mRNA occurs in the areas of somatotopic projection in the lumbar spinal cord. Surprisingly, administration of analgesic doses of morphine or the nonsteroidal antiinflammatory drugs aspirin, metamizol (dipyrone), and indomethacin also increased GPDH mRNA levels in rat spinal cord. The opioid receptor antagonist naloxone completely blocked morphine induction of GPDH but had no effect on GPDH induction by noxious heat stimulation or metamizol treatment, implicating different mechanisms of GPDH induction. Nevertheless, in all cases, induction of the GPDH gene requires adrenal steroids and new protein synthesis, as the induction was blocked in adrenalectomized rats and by cycloheximide treatment, respectively. Our results suggest that the induction of the GPDH gene upon peripheral noxious stimulation is related to the endogenous response to pain as it is mimicked by exogenously applied analgesic drugs.

P-144: Glucose-6-Phosphate Dehydrogenase Activity in Ovine Oocytes in Association with Developmental Characteristics and Expression of Bax Gene

Background: Recent studies have revealed that oocyte competence can be assessed through the presence of the glucose-6-phosphate dehydrogenase (G6PDH) enzyme, as indicated by brilliant cresyl blue (BCB), a dye that can be degraded by G6PDH. In the present study, we aimed to examine the validity of BCB test to select developmentally competent oocyte in ovine and its association with stage-specifi...

Glucocorticoids regulate the transcription of glycerol phosphate dehydrogenase in cultured glial cells.

Utilizing a glycerol phosphate dehydrogenase cDNA clone from the rat glioma C6 cell line, we report that the glucocorticoid regulation of glycerol phosphate dehydrogenase gene expression in glial cells occurs at the transcriptional level. Furthermore, our transcription results in vitro, as well as Northern blot analysis, show that the short-chain fatty acid, sodium butyrate, totally blocks this...

Seasonal freeze resistance of rainbow smelt (Osmerus mordax) is generated by differential expression of glycerol-3-phosphate dehydrogenase, phosphoenolpyruvate carboxykinase, and antifreeze protein genes.

In winter, rainbow smelt (Osmerus mordax) accumulate glycerol and produce an antifreeze protein (AFP), which both contribute to freeze resistance. The role of differential gene expression in the seasonal pattern of these adaptations was investigated. First, cDNAs encoding smelt and Atlantic salmon (Salmo salar) phosphoenolpyruvate carboxykinase (PEPCK) and smelt glyceraldehyde-3-phosphate dehyd...

Transcriptome profiling of brown adipose tissue during cold exposure reveals extensive regulation of glucose metabolism.

We applied digital gene expression profiling to determine the transcriptome of brown and white adipose tissues (BAT and WAT, respectively) during cold exposure. Male C57BL/6J mice were exposed to cold for 2 or 4 days. A notable induction of genes related to glucose uptake, glycolysis, glycogen metabolism, and the pentose phosphate pathway was observed in BAT from cold-exposed animals. In additi...
